Where do I indicate the number of hours an employee is available to work during a day?
.
The number of hours a user is available to work is determined by the company schedule, unless the user has a user-specific work schedule. To create a user-specific work schedule, click on the "Users" link, and then on the name of the user. Click on the "Schedule" link. Select the "User this user-specific work schedule" radio button. Enter the days and number of hours per day the user is available to work, and then click on "Save."

How do I set up my company schedule?
.
The company schedule will affect task lengths. To set up your company schedule, you must have administrative privileges. Go the the My Account module, and select the Company > Schedule tab. You can set the default work days for your company, as well as the number of work hours per day. You can then create any exceptions to this schedule that you need, such as holidays. These exceptions will be taken into effect when the project schedule is created. For example, a task might take additional time if the users assigned to it are not working on a company holiday.

I want to assign a particular user to a task, but
I don't see that user as a choice in the "Assigned to" drop-down menu. Why not?

.
If one or more users have been booked to a project via the Resource module's Bookings tab, *only* those booked users will appear in an associated task's "Assigned to" drop-down menu. You will need to book the particular user to the project in question to be able to assign that user to an associated task, or you must unbook all users currently booked to the project (which will result in all users' names appearing in the task dialog's "Assigned to" drop-down menu).
